A new genus and two new species of Stevardiinae (Characiformes: Characidae) with a hypothesis on their relationships based on morphological and histological data Neotropical Ichthyology

Lepidocharax, new genus, and Lepidocharax diamantina and L. burnsi new species from eastern Brazil are described herein. Lepidocharax is considered a monophyletic genus of the Stevardiinae and can be distinguished from the other members of this subfamily except Planaltina, Pseudocorynopoma, and Xenurobrycon by having the dorsal-fin origin vertically aligned with the anal-fin origin, vs. dorsal fin origin anterior or posterior to anal-fin origin. The present study focused on the effects of aqueous extract of Aegle marmelos (AEAM) on the testis and sperm characteristics induced by cyclophosphamide (CPA) in mice. Due to the recent outbreak of avian influenza, transportation of frozen canine semen with egg yolk has been sharply restricted. Thus, there is urgent need to develop a novel egg yolk-free extender for freezing canine spermatozoa. In the present study, the effect of using skim milk/glucose (SG)-based extender without egg yolk on the motility and fertilizing capacity of canine spermatozoa frozen-thawed in the presence of glycerol was examined. There was a tendency for the proportion of motile spermatozoa exposed to SG-based extender for 3 h to be higher than that exposed for 1 h, but the difference was not significant. Spermatozoa of Natica marochienssish (Gmelin, 1791) is described by light and electron microscopy. The spermatozoon is of the primitive type with head contains a conical acrosomal complex with an acrosomal vesicle of dense matrix having a basis occupied by the subacrosomal space. The middle piece shows the centriolar complex surrounded by mitochondria and the tail contains the axoneme with a 9+2.

Abstract: The objective of this work was to evaluate sperm cell motility after intrafollicular artificial insemination (IFAI) in vivo or after incubation in follicular fluid in vitro. In the in vivo experiment, IFAI was performed, followed by the recovery of follicular content 1 to 4 hours later, in order to assess sperm motility. In the in vitro experiment, spermatozoa from a pool of commercial frozen-thawed semen were evaluated for their kinetics after incubation for 1 or 3 hours, either pure (pool, control group) or in follicular fluid (FF). Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. This study examined the fine structures of epididymal spermatozoa on the lesser white-toothed shrew ( Crocidura suaveolens ), the Japanese white-toothed shrew ( C. dsinezumi ) and the big white-toothed shrew ( C. lasiura ) belonging to the subfamily Crocidurinae living in Korea . In the spermatozoa of C. suaveolens , the head has a large acrosome, a smooth inner acrosomal membrane and a wavy, finger-like, electron-dense apical body. The neck has a solid proximal centriole that is filled with electron-dense material. Resumo: Dez bodes da raca moxoto em idade de 24 a 26 meses foram submetidos durante dois anos a duas colheitas de ejaculados espermaticos semanais em vagina artificial. Os animais foram mantidos em pastagem nativa de caatinga raleada. Na avaliacao quantitativa do semen, encontrou-se maior volume na estacao chuvosa do que na seca. A concentracao de espermatozoides no ejaculado foi inversamente proporcional ao volume, portanto variando com as epocas do ano. Na estacao seca a porcentagem de espermatozoides vivos foi menor do que na estacao chuvosa. ABSTRACT By using informetric tools, we evaluated the contribution and impact of the open source computer-assisted sperm analysis (CASA) software on sperm studies, especially pertaining to fish. The article was cited 125 times between 2007 and 2014 in four publication types (articles, doctoral dissertations, master’s theses, and books). Among articles, the number of citations increased at rate of three citations per year and the calculated h-index was 17. Of the studies that cited Wilson-Leedy and Ingermann (2007), fish were the main group of organisms studied (61 articles); of these, 34 articles used the open source CASA, covering 23 marine or freshwater species. Correlation between sperm characteristics and testosterone in bovine seminal plasma by direct radioimmunoassay R. Bras. Zootec.

The objectives of this study were to validate a non-extractive RIA for seminal testosterone and quantify the hormone using a solid-phase commercial kit, and study the correlation between testosterone in seminal plasma and sperm characteristics. Parallelism showed a correlation index r = 0.992 (Y = -5.47 + 1.073X; R² = 0.985), indicating that the non-extractive method presented is indicated particularly for assessment of testosterone when establishing comparisons between samples. Overall mean (±SD) of testosterone level was 0.60±0.65 ng/mL. Correlation was only found between the seminal concentrations of testosterone and pH of the semen.

Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Determination of optimal glucose concentration for microcalorimetric metabolic evaluation of equine spermatozoa BABT

The heat conduction microcalorimeter can be used to evaluate the metabolic rates of the sperm cell. Two ejaculates of four stallions were cooled to +5ºC and checked for sperm motility (bright field microscopy), viability (eosin 3%), functional membrane integrity (hyposmotic swelling test), and heat production (microcalorimetry). Glucose and sperm cell concentrations were determined in order to measure the heat outputs resulting from sperm metabolism. Sperm viability, membrane integrity and sperm motility did not differ among the different glucose concentrations tested.
